Kisspeptin-10 is a decapeptide: the C-terminal ten residues of the KISS1 gene product, Tyr-Asn-Trp-Asn-Ser-Phe-Gly-Leu-Arg-Phe carried as a C-terminal amide. CAS 374675-21-5, molecular formula C63H83N17O14, molecular weight about 1302. KISS1 was first identified as a melanoma metastasis-suppressor gene (Lee et al., Journal of the National Cancer Institute, 1996), which is where the name metastin comes from.
One molecule carries three names, all of which appear on vendor pages and in trial registrations. Metastin 45-54 counts the same residues against the 54-amino-acid peptide the ligand was first isolated as, and kisspeptin-112-121 counts them against the 138-amino-acid KISS1 precursor, which is how Massachusetts General Hospital registers the compound on ClinicalTrials.gov.
Kisspeptin-10 agonizes KISS1R, also called GPR54, a class-A GPCR on hypothalamic GnRH neurons that couples through Gq/11. A 2024 cryo-EM study in Cell Reports (Shen et al.) solved KISS1R bound to kisspeptin-10 and to the synthetic analog TAK-448, and found both share a binding mode built on the receptor's extracellular loops.
That places it a rung above the compounds it is shelved with. Kisspeptin drives GnRH release, whereas gonadorelin is GnRH itself, acting on the pituitary. hCG skips both and agonizes the LH receptor on the gonad directly. The upper two rungs need an intact pituitary to produce anything and the third does not.
The IVF work at Imperial College London used kisspeptin-54: it triggered egg maturation in 53 women undergoing IVF, with fertilization and embryo transfer in 49 of them (92%) and clinical pregnancy in 12 (23%) (Jayasena et al., Journal of Clinical Investigation, 2014). So did the same group's brain-imaging and hypoactive-sexual-desire studies. The two lengths behave differently in vivo: in male mice, KP-54 had a plasma half-life near 32 minutes against roughly 4 minutes for KP-10, repeated KP-10 injections failed to reproduce the sustained LH rise a single KP-54 injection produced, and only KP-54 activated c-FOS in GnRH neurons behind the blood-brain barrier (d'Anglemont de Tassigny et al., PLoS One, 2017).
The decapeptide does have its own clinical literature. George et al. (Journal of Clinical Endocrinology and Metabolism, 2011) gave intravenous kisspeptin-10 to healthy men and recorded a rapid LH rise, from 4.1 to 12.4 IU/L at thirty minutes (group means in six men at the most effective bolus dose). In four men, a 22.5-hour infusion raised mean LH from 5.4 to 20.8 IU/L and testosterone from 16.6 to 24.0 nmol/L, while a lower rate raised LH pulse frequency from 0.7 to 1.0 pulses per hour. A 2013 follow-up in Clinical Endocrinology found a comparable LH rise in five men with type 2 diabetes and low testosterone, and Massachusetts General Hospital has run a long program under the kisspeptin 112-121 label, including a 256-participant study completed in 2021.
Industry pursued longer-acting analogs instead. MVT-602 produced an LH rise of similar amplitude to kisspeptin-54 in nine healthy women but peaked at 21.4 hours instead of 4.7, with roughly four times the area under the LH curve (Abbara et al., Journal of Clinical Investigation, 2020). Takeda's TAK-448 reached Phase 2 in hypogonadal men, and both Phase 2 studies were terminated, in November 2015 and April 2016, each for failing its primary endpoint.
