FOXO4-DRI

A cell-penetrating D-retro-inverso peptide (CAS 2460055-10-9) derived from the FOXO4 forkhead domain, in which every residue is a D-amino-acid and the sequence is reversed — an architecture that preserves three-dimensional binding geometry while conferring resistance to proteolysis. Competes with endogenous p53 for binding to FOXO4, releasing p53 to mitochondria and triggering apoptosis selectively in senescent cells. Reported in the 2017 Baar Cell paper to extend median lifespan 24.8% in naturally aged mice and restore fur density, renal function, and fitness. Preclinical-stage senolytic candidate.
