Testagen is a synthetic tetrapeptide, Lys-Glu-Asp-Gly (written KEDG), from Vladimir Khavinson's short-peptide program, the family that also produced Epitalon, Pinealon and Cartalax. PubChem's record for H-Lys-Glu-Asp-Gly-OH gives the formula C17H29N5O9 and a molecular weight of about 447. Two reviews by Khavinson and colleagues list Testagen as a regulator of the male reproductive system, one in Molecules in 2021 and one in the International Journal of Molecular Sciences in 2022. The 2022 table cites no source for that entry. The 2021 table cites three papers: a study of the peptide binding plant histones, a review of peptide-driven cell differentiation, and a study of two other peptides, Vilon and Epitalon, in mouse heart. That differentiation review lists KEDG among peptides that stimulate immune-cell differentiation (Khavinson, Stem Cell Reviews and Reports, 2020).
A PubMed search for the name and the sequence in October 2026 found no study of the peptide in testicular or other reproductive tissue.
The animal work on the sequence comes from a group at Chita State Medical Academy in Russia. Its papers describe Lys-Glu-Asp-Gly as synthesized from the amino acid composition of peptide extracts of the anterior pituitary, and test it alongside Ala-Glu-Asp-Gly, the Epitalon sequence, which they trace to the posterior lobe. The two differ only in their first residue.
The same group's 2008 Russian patent, RU2325397, claims Lys-Glu-Asp-Gly as a medicine that stimulates thyroid-stimulating hormone and thyroid hormone production.
The family's proposed mechanism is direct binding to DNA and histones. For Testagen, a fluorescently labelled copy entered the nuclei of cultured HeLa cells, and fluorescence measurements suggested it binds short DNA strands, preferring sequences containing CAG (Fedoreyeva, Biochemistry (Moscow), 2011). It also bound wheat histones H1, H2B, H3 and H4 (Fedoreyeva, Biochemistry (Moscow), 2013), and computer docking by Khavinson and colleagues predicts it can bind the LAT1, LAT2 and PEPT1 transporters (Biomolecules, 2023). The papers name no receptor for it.
