A 21- or 24-amino-acid mitochondrial-derived peptide (CAS 330936-69-1) encoded in a small open reading frame within the 16S ribosomal RNA gene (MT-RNR2) of mitochondrial DNA. Discovered in 2001 via cDNA screening of surviving neurons in Alzheimer's disease brain; the prototype of the mitochondrial-derived peptide class (which also includes MOTS-c and SHLP1-6). Cytoprotective, anti-apoptotic, and insulin-sensitizing activity demonstrated preclinically in models of Alzheimer's disease, ischemia, and metabolic dysfunction. Circulating levels decline with age.
